Provision of the Centre of Collective Use

1. 1. Creation of the Centre
1.1. 1.1. Centre of collective use of the device cytofluorometer EPIC® XLTMFlow Cytometer and Mass-Spectrometer Voyager-DE TM PRO Biospectrometry TM Workstation (далі - ЦККП) of NAS of Ukraine is created on the basis of the Institute of Biochemistry of NAS of Ukraine with the aim of the most rational use of a unique and expensive research device of foreign production which acquisition is impossible or inexpedient for any scientific institution for the centralized assets, common assets of groups, institutions and organizations, other financing sources of the National Academy of Sciences of Ukraine.
1.2 1.2. The Institute of Biochemistry of NAS of Ukraine undertakes the obligation to ensure a reliable and high-quality operation of cytofluorometer EPIC® XLTM Flow Cytometer and Mass-Spectrometer Voyager-DE TM PRO Biospectrometry TM Workstation, its necessary special maintenance, conditions of the best use of the working hours in the interests of scientific community of NAS of Ukraine, absence of artificial restrictions in rendering services to other scientific institutions and organizations of NAS of Ukraine.

2. Structure, main tasks and work organization of the Centre
2.1. Centre of collective use of the device cytofluorometer EPIC® XLTMFlow Cytometer and Mass-Spectrometer Voyager-DE TM PRO Biospectrometry TM Workstation is a property of the National Academy of Sciences of Ukraine. Cytofluorometer is on the balance of the Institute of Biochemistry of NAS of Ukraine, the place of the Centre creation.
2.2. CCUD of NAS of Ukraine is a research subdivision of the Institute of Biochemistry of NAS of Ukraine subordinate to the Institute directorate without a status of legal person. CCUD of NAS of Ukraine is called “Centre of Flow Cytometry and Mass-Spectrometry”.
2.3. The main task of CCUD at the Institute of Biochemistry of NAS of Ukraine is providing the researchers of NAS of Ukraine with the possibilities to conduct researches with cytofluorometer EPICS® XLTM Flow Cytometer and Mass-Spectrometer Voyager-DE TM PRO Biospectrometry TM Workstationmaintained by highly qualified personnel capable to keep equipment in good serviceable condition and to render advice services.
2.4. General management of the work of CCUD of NAS of Ukraine is given the deputy director in research work of the Institute of Biochemistry of NAS of Ukraine.
2.5. The Institute of Biochemistry of NAS of Ukraine informs (no less than once a year) the Bureau of the Department of MBBECD of NAS of Ukraine about the work of the Centre, which is its part, and, if is necessary, the Commission in the problems of modernization of the park of research devices and equipment at NAS of Ukraine (hereinafter referred to as the Commission). The general annual report includes the data on the work of CCUD of NAS of Ukraine which are submitted by an Institution of NAS of Ukraine, as a separate section on paper and an electronic version, the Presidium of NAS of Ukraine, Bureau of the MBBECD department of NAS of Ukraine and to the Commission. It should be noted that the standard structure of such a section is developed by the Commission.
2.6. The Bureau of the MBBECD Department of NAS of Ukraine considers at its meetings the questions of the work of CCUD of NAS of Ukraine which function at its institutions. If the Centre work is nonsatisfactory the Department can rise a question about the change of the headquarters scientific institutions and equipment transfer to this institution, or the Centre closing..

3. Order of rendering services. 3.1. Information about CCUD of NAS of Ukraine (type/grade of research device/equipment, presented for collective use, its main technical data and main trends of researches which may by performed, using such equipment) may be found on the web-page of the Institute www.biochem.kiev.ua. Such information about all the CCUD of NAS of Ukraine and their basic institutions at NAS of Ukraine may be found on the web-page of the Presidium of NAS of Ukraine.
3.2. Scientific institutions and organizations of NAS of Ukraine, which have a necessity in performing researches using scientific equipment of CCUD of NAS of Ukraine (hereinafter referred to as Customers) would make applications in writing to put them in Bureau of MBBECD Department. The application includes the number of hours, terms and kinds of investigations which performance needs using the research devices and equipment of CCUD. The applications are made twice a year before January 15 and June 15. 3.3. It should be taken in consideration that the Bureau of the MBBECD Department of NAS of Ukraine, on presentation of Director of the Institute of Biochemistry of NAS of Ukraine, approves general distribution of working hours allowed for the collective use of the Centre research equipment.
3.4. Director of the Institute of Biochemistry of NAS of Ukraine defines, by his order agreed with the Bureau of MBBECD Department, the operation schedule of the Centre and certain research devices/equipment, established the required number of operation shifts per a working day with allowance for the operation schedule of the scientific institution and needs of researchers of NAS of Ukraine in using the research device/equipment.
3.5. Under one-shift operation (8 hours of working time) 3 hours are given for the Institute needs, 3 hours free of charge are given the customers – other scientific institutions and organizations of NAS of Ukraine, 2 hours are left for the paid use of the Centre equipment by other institutions or enterprises and organizations which are not under the authority of NAS of Ukraine in accordance with Law in force. Under the two-shift operation (12 hours of working time) the time distribution corresponds to the ratio 5:4:3. The working hours of research devices/equipment are recorded in the operation logs, the record form being defined by the Commission.
3.6. When planning the use of working hours of the research device Director of the Institute may define other ratios of the use of working hours for the Institute needs and paid services, but the time of the collective free use of the centre research devices remains unchanged, i.e., 3 hours at 8-hour working day and 4 hours at 12-hour working day.
3.7. Paid services when using research equipment of CCUD of NAS of Ukraine in accordance with demands of other customers are rendered due to current legislation of Ukraine. 3.8. The Institute offsets all costs connected with the use of research equipment presented by CCUD of NAS of Ukraine.

4. Change of the headquarters institution, closing of the Centre 4.1. It should be taken in consideration, that in case of nonfulfilment of requirements of the given standard Provision, lack of the corresponding organization of the work of CCUD of NAS of Ukraine on the part ob the Institute in providing collective use of CCUD equipment for requirements of researchers at NAS of Ukraine or arising of other force-majeure circumstances the Bureau of MBBECD of NAS of Ukraine submits proposals for the Commission consideration concerning the transfer of the equipment assigned to CCUD of NAS of Ukraine, to another research institution of NAS of Ukraine with corresponding change of the Centre name and its location.
4.2. The Commission considers the proposals of MBBECD Department of NAS of Ukraine in accordance with sections 1 and 2 of the given Provision, takes decisions on the change of headquarters research institution or on closing the CUUD of NAS of Ukraine and submits it for approval of the Presidium of NAS of Ukraine.

The Flow Cytofluorometer COULTER® EPICSTM XLTM Flow Cytometer is a system for high-quality and quantitative determination of biological and physical properties of cells and other particles. These properties are measured when cell are passing through the laser beam. The device can help to measure 6 parameter at once: forward scattering (FS), side scattering (SS) and fluorescence intensity of particles at 4 wavelengths (FL1, FL2, FL3, FL4), using 1 laser which radiates at wavelength of 480 nm. Then the device can make the correlation multiparameter analysis of separate cells. Using the device, one can investigate such biological objects of human, animal and plant cells, sea plankton, bacteria, etc.

   Set of equipment for cytofluorometric analysis:

  1. Cytofluorometer COULTER® EPICS® XLTM Flow Cytometer - analyzes biologic samples, requires 2 solutions for an operation:  A.ISOFLOWTM - transparent buffer solution which does not contain particles greater than 0.2 мкм, is characterized by low background, bacteriostatic and fungistatic properties, is in optimal correspondence by pH, osmolarity, conductivity to characteristics of biologic samples analyzed.
    B.COULTER CLENZ® - washing solution which permit cleaning the device parts which contact with biological sample from proteins and cells adherence.
  2. Preparation work station Q-PREPTM WORKSTATION - permits making blood preparations in automatic conditions for investigating by cytofluorometer, i.e. to make lysis of erythrocytes, fixation of cells and their staining with fluorescence-labeled antibodies.
  3. Electric power supply source – provides electric power supply, pressure and vacuum for cytofluorometer and includes a capacity of 4 l in volume for gathering waste.
  4. Computer – controls flow cytometer work, stores obtained data; it consists of the system unit, monitor, “mouse”, keyboard.
  5. Printer HP 1600C color printer – permits listing the measurement results in a form of black-white and coloured histograms.

Specifications of electric power supply and operation condition:
There are two necessary separate lines of 115-V and 20-A with earth. The room temperature should be of The room temperature should be of 16oC – 32oC, and its shifts should not exceed 30C an hour. ПAdmissible air humidity from 5% to 80%. The air should be permanently cooled, since the heat scattering under the device operation is 2 300 W.

Mass-spectrometr Voyager-DE TM PRO Biospectrometry TM Workstation is a biospectrometrical system for identification and characteristic of proteins and peptides in the prodark analysis. This biospectrometrical system uses МАLDI-TOF technology in linear and reflective variants, for obtaining concrete data about weights of studied substances in broad spread of weight. The working station Voyager possesses all advantages of systems with matrix-bound laser ionization (МАLDI) and Time-of-Flight detection for ensuring investigation problems (but not for diagnosis purposes). Peptides and proteins with m.w.?10000 Da, small oligonucleotides with m.v. ?3500 Da, acid glyceropeptides, large oligonucleotides with m.v.? 3500 Da, neutral and alkali carbohydrates, glycolipids, polar and nonpolar synthetic polymers. Aromatic polymers, nonpolar polymers, small molecules may be investigated in the system МАLDI-TOF with availability of matrices (substances capable to initiative ionization process) using biospectrometer Voyager-DE TM PRO Biospectrometry TM Workstation.

1. Working station Voyager, is made of:

- One-level reflector with optimized optics for PSD- analysis;
- Horizontal flight tube (linear regime -1.3 m, reflected regime - 2.0).
- Two-level source of ions, voltage of ion source may be regulated: accelerating voltage up to 25000 V and voltage on the control grid (the range is determined by accelerating voltage)
- Nitrogen laser, 337 nm, pulse time 3 ns, power 36 mW, with maximum frequency 20 Hz, with air cooling, controlled by the computer program. Laser frequency is regulated by the converter GHz for increasing potentialities.
- Vacuum system consists of 1 forevacuum and 2 turbomolecular pumps for double differentiation of turbomolecular suction for obtaining deep vacuum, and multivacuum control-measuring capacity for independent monitoring of the source ionizer regions.
- Automatic system for changing the sample. Possibility to be used for applying the plate specimens of different format, manual control with the help of computer joystick or mouse, software for automatic analysis.
